Read these instructions carefully before using your SEMGLEE. If you are not able to follow all the instructions completely on your own, use SEMGLEE only if you have help from a person who is able to follow the instructions.

People who are blind or have vision problems should not use SEMGLEE single-patient use prefilled pen without help from a person trained to use SEMGLEE single-patient-use prefilled pen.

Follow these instructions completely each time you use SEMGLEE to ensure that you get an accurate dose. If you do not follow these instructions you may get too much or too little insulin, which may affect your blood glucose.

SEMGLEE is a disposable pen for the injection of insulin. Each SEMGLEE contains in total 300 units of insulin. You can set doses from 1 to 80 units in steps of 1 unit. The pen plunger moves with each dose. The plunger will only move to the end of the cartridge when 300 units of insulin have been given.

    Carcinogenesis Mutagenesis Impairment Of Fertility

    In mice and rats, standard two-year carcinogenicity studies with insulin glargine were performed at doses up to 0.455 mg/kg, which was for the rat approximately 10 times and for the mouse approximately 5 times the recommended human subcutaneous starting dose of 10 Units/day , based on mg/m . The findings in female mice were not conclusive due to excessive mortality in all dose groups during the study. Histiocytomas were found at injection sites in male rats and male mice in acid vehicle containing groups. These tumors were not found in female animals, in saline control, or insulin comparator groups using a different vehicle. The relevance of these findings to humans is unknown.2

    Insulin glargine was not mutagenic in tests for detection of gene mutations in bacteria and mammalian cells and in tests for detection of chromosomal aberrations .

    In a combined fertility and prenatal and postnatal study in male and female rats at subcutaneous doses up to 0.36 mg/kg/day, which was approximately 7 times the recommended human subcutaneous starting dose of 10 Units/day , based on mg/m , maternal toxicity due to dose-dependent hypoglycemia, including some deaths, was observed. Consequently, a reduction of the rearing rate occurred in the high-dose group only. Similar effects were observed with NPH insulin.2

    Warnings For Other Groups

    For pregnant women: It is not known if insulin glargine is safe to use in pregnant women.

    Tell your doctor if youre pregnant or plan to become pregnant. You should only use insulin glargine during pregnancy if the potential benefit justifies the potential risk.

    For women who are breastfeeding: It isnt known if insulin glargine passes into breast milk. You and your doctor may need to decide if youll use insulin glargine or breastfeed. If you do both, your insulin glargine dosage may need to be adjusted, and your blood sugar level may be closely monitored.

    For seniors: People aged 65 years or older may be more sensitive to insulin glargine. This may raise your risk of a low blood sugar reaction. You doctor may start you on a lower dosage, and increase your dosage slowly.

    For children: Talk to your childs doctor about the use of insulin glargine in children. Special care may be needed.

    Why Is This Medication Prescribed

    Insulin glargine products are used to treat type 1 diabetes . Insulin glargine products are also used to treat people with type 2 diabetes who need insulin to control their diabetes. In people with type 1 diabetes, insulin glargine products must be used with another type of insulin . In people with type 2 diabetes, insulin glargine products also may be used with another type of insulin or with oral medication for diabetes. Insulin glargine is a long-acting, manmade version of human insulin.     The dose of this medicine will be different for different patients. Follow your doctor’s orders or the directions on the label. The following information includes only the average doses of this medicine. If your dose is different, do not change it unless your doctor tells you to do so.

    Did you know your blood sugar can be too low as well as too high? Those swings may happen with insulin treatment. Just know that monitoring your blood sugar as directed by your doctor gives you useful information that can help manage your diabetes.1

    CHANGES ARE NORMAL

    Things like stress, your diet, other medications, exercise, and more can affect your blood sugar. This is completely normal.     STAY ALERT FOR LOW BLOOD SUGAR

    How To Choose An Injection Site

    Insulin should be injected into the fatty tissue just under your skin. Avoid injecting insulin into your muscle. Choose one of the following injection sites :

    • Your abdomen , except for the 2-inch area around your belly button. If youre injecting rapid-acting insulin, this is the best site to use.
    • The upper and outer part of your thighs. If youre injecting long-acting insulin, this is the best site to use.
    • The upper outer part of your buttocks .
    • The back part of your upper arms.
    • Only use this injection site if someone else is giving you the injection. If youre giving yourself the injection, its too hard for you to reach the correct area at the back of your arm.

    Keep track of the injection site you use. Be sure to rotate the injection sites with each injection. Within each injection site, always inject 1 to 2 inches away from the last place you injected. This can help you avoid soreness and scar tissue.

    Make sure to inject at least 2 inches away from any incisions , scars, or stretch marks.

    Dont inject into an area thats tender, red, bruised, or hard.

    Dosage For Type 1 Diabetes

    If you have type 1 diabetes, the starting dose of Lantus is generally about one-third of your total daily insulin dose.

    Your total daily insulin dose is based on your weight. It typically ranges from 0.4 to 1.0 units per kilogram a day.

    For example, a 150-lb man weighs about 68 kg. If his doctor prescribes 0.5 units/kg a day, his total daily insulin dose would be 34 units a day. His starting dose of Lantus would be about one-third of 34 units, which is about 11 units of Lantus.

    Lantus is typically taken once a day, not twice a day.

    The drug is given as an injection just under your skin . You can inject Lantus in the skin of your upper arms, belly , or thighs.

    Avoid medication errors by always checking the medicine label before injecting your insulin.

    What Medications Are Similar

    Lantus contains the ingredient insulin glargine. Insulin glargine is also available as other brand-name insulins:

    • Basaglar: Contains insulin glargine and is available in the same dosage of Lantus, which is 100 units per mL. Basaglar is indicated for use in adults with type 1 or type 2 diabetes and in children at least 6 years old with type 1 diabetes.
    • Semglee: A biosimilar product to Lantus. Semglee contains 100 units per mL insulin glargine, the same dosage as Lantus. Semglee is the first interchangeable biosimilar insulin product approved by the FDA. This means that Semglee can be substituted for the brand-name drug Lantus. Semglee is for use in adults with type 1 or type 2 diabetes and in children at least 6 years old with type 1 diabetes.
