Total RNA was extracted from liver or adipose tissue using a Sigma TRI reagent kit, according to the manufacturer’s protocol, and was then dissolved in diethylpyrocarbonate-treated water. For cDNA synthesis, total RNA was reverse transcribed using PowerScript RT , according to the protocol supplied, using a combination of oligo15 primer and random hexamers. PCR was performed using the GoTaq®Green Master Mix PCR kit in a 20 l reaction mixture containing 1 l of the RT reaction mixture and 0·5 l of each primer . PCR primers were designed using published nucleotide sequences for GCK, G6Pase, PEPCK, GLUT2, PPAR-, PPAR-, GLUT4, SREBP-1c and -actin.

PCR using the GCK primer was performed with an initial cycle of 4 min at 94°C, followed by twenty-two cycles of 30 s at 94°C, 30 s at 57°C, and 30 s at 72°C, and a final extension for 5 min at 72°C. PCR using the G6Pase, PEPCK, PPAR-, PPAR-, GLUT2, GLUT4, SREBP-1c and -actin primers was performed similarly, with the exception of the annealing temperature and the number of cycles . The -actin transcripts were used as internal controls.
